In response to your letter in which you ask about the vowels according to the teachings of Chachmei HaNistar (Kabbalah): Regarding their sanctity—not only the sanctity of the vowels themselves but even the sanctity of their names—it is understood from what is written in Tikunei Zohar, Tikkun 5, in the introduction (7b), Tikkun 69, Zohar Chadash (99c) in the Tikkunim, that even their names are acronyms for angels.

Regarding their gematria: it is straightforward—each dot (pintele) is ten, and each line is six. Therefore, for example, Tzeirei ( ֵ ) is twenty, and similarly for others. For example, see Tikunei Zohar in the introduction, where Kamatz has a gematria of sixteen, and so on.

I am enclosing something recently published which you will surely study as it fits this topic.

Sometimes the vowels are calculated according to their name—as in the introduction to Tikunei Zohar there (7b): Patach—when counting its letters—is yud-vav-daled; as explained by commentators, the letters of this vowel's name have a gematria of twenty (in small numbers). For example: Peh = 80 (small number: 8), Tav = 400 (small number: 4), Chet = 8; so 8+4+8=20. And when written as yud-vav-daled =20 (yud=10, vav=6, daled=4=20).
